Amlodipine is one of the first-line antihypertensive drugs available in tablet form which has shortcomings, especially for patients who have difficulty swallowing the drug, so it was developed in the form of ODT preparations. Oral Disintregant Tablets (ODT) are tablet preparations that can be destroyed in less than 60 seconds in the mouth. An important component in the ODT preparation formula is the crushing agent. In this study, the crushing agent Sodium Starch Glycolate (SSG) and Avicel pH 102 were used. SSG is a superdisintegrator with a swelling working mechanism. Avicel pH 102 can function as a crushing agent that uses a capillary action mechanism (wicking action). The preparation of this ODT preparation uses the active ingredient Amlodipine Besylate and a combination of Sodium Starch Glycolate (SSG) and Avicel pH 102 crushing agents with consecutive concentration presentations of F1 (4:0), F2 (2:5), and F3 (2:2.5) using the wet granulation method. This study aims to determine the influence of the use of a combination of crushing materials and to find out what is the good composition in the manufacture of Amlodipine ODT. With the results of the destruction time test obtained the average and standard deviation in F1, F2, and F3 respectively, which are 38.3 ± 0.471; 51.0 ± 0.816; and 31.3 ± 1.247. Based on these results, it can be concluded that the three formulas are found to have the influence of a combination of crushing materials.
