ABSTRAK Loratadine HCl merupakan obat antihistamin golongan reseptor H1 yang digunakan untuk pengobatan alergi. Loratadine HCl tersedia dalam bentuk tablet 10 mg. Pembuatan OFDT loratadine HCl dalam formulasi ini untuk mengetahui mana hasil yang lebih optimal dari perbedaan konsentrasi bahan penghancur Sodium Starch Glycolate dengan konsentrasi 2%, 5%, dan 8%. Oral Fast Dissolving Tablets (OFDT) merupakan tablet sediaan obat padat yang mudah hancur bila diletakkan diatas lidah dan dapat memberikan efek terapi yang cepat. Tujuan : Peneilitian ini bertujuan untuk mengetahui konsentrasi Sodium Starch Glycolate (SSG) terhadap mutu fisik sediaan Oral Fast Dissolving Tablets merupakan superdisintegrant yang dapat digunakan sebagai bahan penghancur. Metode : Penelitian eksperimental dan menggunakan metode granulasi basah. Hasil : Pembuatan Oral Fast Dissolving Tablets Loratadine HCl menggunakan metode granulasi basah dengan variasi konsentrasi Sodium Starch Glycolate 2%, 5% dan 8% untuk uji keseragaman bobot, keseragaman ukuran, kekerasan tablet dan keseragaman kandungan telah memenuhi persyaratan dan uji waktu hancur konsentrasi 2% dan 5% memenuhi persyaratan tetapi untuk konsentrasi 8% tidak memenuhi persyaratan. Data uji mutu fisik yang dihitung dengan One Way Anova yang menunjukkan nilai signifikan lebih dari 0,05 tidak ada perbedaan Sodium Starch Glycolate 2%, 5% dan 8% terhadap mutu fisik Oral Fast Dissolving Tablet loratadine HCl. Dalam pembuatan Oral Fast Dissolving Tablet semakin rendah konsentrasi dari Sodium Starch Glycolate maka dapat memberikan hasil mutu yang baik. Moreover, the manufacture of OFDT loratadine HCl in this formulation is to find out which results are more optimal than the different concentration of the disintegrant Sodium Starch Glycolate with concentration of 2%, 5%, and 8%. Furthermore, Oral Fast Dissolving Tablets (OFDT) is a solid drug dosage form which dissolves easily when it is placed on the tongue; besides, it can provide rapid therapeutic effect. Purpose: This study aims to determine the concentration of Sodium Starch Glycolate (SSG) on the physical quality of Oral Fast Dissolving Tablets. Sodium Starch Glycolate is a superdisintegrant which can be used as a disintegrant. Method : Experimental research and using the wet granulation method. Results : Manufacture of Oral Fast Dissolving Tablets Loratadine HCl using wet granulation method with various concentrations of Sodium Starch Glycolate 2%, 5% and 8% for the test of weight uniformity, size uniformity, tablet hardness and content uniformity has met the requirements and the disintegration time test for concentrations of 2% and 5% met the requirements but for a concentration of 8% did not meet the requirements. Physical quality test data calculated by One Way Anova which shows a significant value more than 0.05 no difference Sodium Starch Glycolate with concentration of 2%, 5%, and 8% on the physical quality of Oral Fast Dissolving Tablets Loratadine HCl. In the making Oral Fast Dissolving Tablets, the lower concentration of Sodium Starch Glycolate then it can give good quality results.
